  • Abstract: Oak tasar silk moths were collected from different regions of North East States. Tissues were excised by a sterile blade and stored by dipping in RNA Later solution at minus 80 degree .Genomic DNA was extracted from the received silk worm tissues using a CTAB reagent-based method. The quality and quantity of the extracted DNA samples were assessed using a NanoDrop and subjected to agarose gel electrophoresis.A 16S microbiome profiling was conducted on the MiSeq platform with a read length of 2x300 bp. The following samples were analysed: Antheraea proylei RSRS Imphal APM,Antheraea proylei C27 APC2, Antheraea proylei Mizoram APMi, Antheraea proylei Senapati APS, Antheraea proylei Chandel APC, Antheraea pernyi RSRS Imphal Aper, Antheraea roylei Senapati AR, Antheraea frithi Senapati AF.The extracted metagenomic DNA was amplified using a bacterial 16S V3-V4 region-specific primer set. Three microlitres of the PCR product was resolved on a 1.2 percent agarose gel at 120V for approximately 60 minutes or until the sample reached 3 by 4th of the gel. 16S metagenomic comparative profile indicates a diverse but Firmicutes Proteobacteria-dominated microbiota. Moderate levels of Actinobacteriota and Bacteroidota imply stable metabolic activity, whereas minor phyla add ecological complexity.The rarefaction analysis suggests adequate sequencing coverage and reveals that AR and AF possess the most diverse microbiota, possibly reflecting more complex or metabolically active environments. In contrast, Aper and APM have relatively fewer taxa, indicating lower community richness-potentially due to environmental stress, and physiological stage.
