- Descriptive Title: A thermotolerant isolate, Saccharomyces cerevisiae NGY10, obtained from sugarcane distillery waste, exhibits high potential for lignocellulosic ethanol production. The strain is capable of efficient fermentation at 40 degree Celsius, achieving 92.81 and 86.43 percent fermentation efficiency during separate hydrolysis and fermentation and simulatenous saccharification and fermentation processes , respectively, using dilute acid-pretreated rice straw. The genome of NGY10 was sequenced using hybrid approach combining Illumina and Oxford Nanopore technologies. De novo assembly resulted in 39 contigs, with a total genome size of 12.8 Mb.

- Abstract: In this study, we report the hybrid de novo genome assembly of Saccharomyces cerevisiae isolate NGY10, a robust yeast strain isolated from sugarcane distillery waste. Saccharomyces cerevisiae NGY10 exhibits desirable traits for lignocellulosic bioethanol production, including thermotolerance at 40 degree Celsius and resistance to key fermentation inhibitors - furfural, 5-hydroxymethylfurfural , acetic acid and ethanol. Using a combination of Illumina and Nanopore sequencing, a high quality hybrid genome assembly was generated, consisting of 39 contigs with a total length of 12.96 Mb, a maximum contig size of 1.76 Mb and N50 Value of 603,771 bp and no ambiguous bases. The high quality genome of NGY10 provides a valuable resource or future metabolic engineering aimed at enhancing thermotolerance, inhibitor resistance, and lignocellulosic utilization for sustainable bioenergy production.
