- Abstract: Genomic DNA was isolated from leaves using ArborEasyŽ DNA isolation kit (ICFRE-Institute of Forest Genetics and Tree Breeding, Coimbatore, India). DNA concentration and purity was estimated using NanoDrop ND-1000 UV-Vis Spectrophotometer (Thermo Scientific, MA, USA) and DNA was quantified using Qubit Fluorometer (Thermo Scientific, MA, USA). Library construction was conducted using KAPA HyperPlus Kit (Roche Sequencing Solutions Inc., Pleasanton, CA, USA) to generate fragments of size of 300-500bp. The library was subsequently quantified using Qubit Fluorometer (Thermo Scientific, MA, USA) and size distribution was checked on Agilent 2100 Bioanalyzer (Agilent Technologies, USA). The library was sequenced on Illumina NovaSeq (Illumina Inc., San Diego, CA, USA) using 150 bp paired-end chemistry.
